Antoine Barrière and Amel Toudji-Zouaz in the Bertrand team developed a new method to image and quantify transcription in vivo.

To understand how cells acquire their identity during development, it is essential to follow gene expression dynamics in vivo. Antoine Barrière and Amel Toudji-Zouaz in the Bertrand team developed a new method to image transcription using the Argonaute protein NRDE-3 tagged with a fluorescent protein. NRDE-3 is programmed with dsRNA targeting a gene of interest and labels nascent transcripts at transcription sites and mRNA in the cytoplasm. In C. elegans, dsRNA are easily introduced by feeding the animals. Expression of the gene of interest is then followed and quantified over time by in vivo fluorescent microscopy.
